Target Information
Junctional complexes between the plasma membrane (PM) and endoplasmic/sarcoplasmic reticulum (ER/SR) are a common feature of all excitable cell types and mediate cross talk between cell surface and intracellular ion channels.
Junctophilins (JPs) are essential components of these complexes, composed of a hydrophobic segment spanning the ER/SR membrane and a cytoplasmic domain with affinity for the PM.
Four tissue-specific subtypes are identified:
- JPH1: Skeletal muscle
- JPH2: All muscle cell types
- JPH3, JPH4: Predominantly expressed in the brain
In the CNS, JPH3 and JPH4 contribute to subsurface cistern formation in neurons. Mice lacking both subtypes exhibit impaired learning, memory, and abnormal nervous functions.
