Target Information
FGF2 (also known as FGFb, fibroblast growth factor basic) is a member of the fibroblast growth factor (FGF) family. It interacts with high-affinity transmembrane receptors to regulate cell proliferation and tissue neovascularization. FGF2 exists as five isoforms with distinct intracellular localizations and functions.
- The 18 kDa isoform is cytosolic and acts through cell surface receptors.
- The 22, 22.5, 24, and 34 kDa isoforms are nuclear and may signal independently of transmembrane receptor pathways.
In humans, the FGF2 gene is located on chromosome 4q. It plays roles in limb and nervous system development, wound healing, and tumor growth. The mRNA of FGF2 contains multiple polyadenylation sites and is alternatively translated from non-AUG and AUG initiation codons, producing five isoforms with distinct properties.
CUG-initiated isoforms localize to the nucleus and mediate intracrine signaling, while AUG-initiated isoforms are cytosolic, mediating paracrine and autocrine effects.
FGF2 dysfunction is associated with diseases such as Kaposi Sarcoma and corneal neovascularization.
