Target Information
FOXO1 (FKHR, Forkhead Box O1)는 전사 인자(forkhead homeotic gene family)의 하위군에 속하는 단백질로, 핵 수용체 활성을 조절하는 공활성자(coactivator) 또는 공억제자(corepressor)로 작용할 수 있습니다.
FOXO1은 PAX3와의 결합을 통해 alveolar rhabdomyosarcoma에 관여하며, 항암 활성, 자가포식(autophagy), 근육 성장 및 분화 과정과도 관련이 있습니다.
이 단백질은 인슐린 신호 전달의 주요 표적이며, 산화 스트레스에 대한 대사 항상성을 조절합니다. 또한 FOXO1은 redox 균형, 골아세포 수 조절, 골량 조절에 관여하며, 골격의 내분비 기능을 통해 포도당 대사를 조절합니다.
간세포에서는 PPARGC1A와 함께 작용하여 IGFBP1, G6PC, PCK1 등의 유전자 발현을 활성화함으로써 포도당신생(gluconeogenesis)을 촉진합니다. FOXO1은 CDK1, PKB/AKT1, SKT4/MST1 등의 신호 하류에서 세포 사멸을 조절하는 중요한 인자입니다.
