Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein, which exists in two isoforms (345 and 395 kDa) expressed in dividing cells. Ki-67 is present during active phases of the cell cycle (G1, S, G2, and M) but absent in resting cells (G0). During interphase, expression is localized to the nucleus and redistributes to chromosomes during mitosis, associating with heterochromatin-bound proteins such as CBX3.

In tumor studies, Ki-67 serves as a marker for determining the fraction of proliferating cells within a tumor population. This clone also recognizes canine Ki-67.

Applications Reported: Intracellular staining using Foxp3 Staining Buffers followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested with Foxp3/Transcription Factor Buffers Set (Product #00-5523-00) on normal human peripheral blood cells.

Refer to BestProtocols® for detailed intracellular staining procedures (Protocol B: One-step protocol for intracellular/nuclear proteins).
Recommended usage: 5 µL (0.03 µg) per test in a final volume of 100 µL. Cell number range: 10⁵–10⁸ cells/test.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with a 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable).

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal signal loss. Clone-specific performance should be verified empirically.

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It contains a forkhead-associated (FHA) domain that enables association with euchromatin and nucleolar regions. Ki-67 distribution varies by cell cycle phase: perinucleolar (G1), nuclear matrix (G2), and chromosomal (M phase).
As a proliferation marker, Ki-67 is absent in G0 cells and increases through mitosis. It is widely used to assess the proliferative index in neoplasms, with strong correlation between low Ki-67 index and low tumor grade. It is also used as a neuronal marker of cell cycling and proliferation.
