  • Specificity Comments:
    Osteopontin is a bone matrix protein and cytokine secreted by activated T cells. It is an extracellular matrix cell adhesion phosphoglycoprotein deposited into unmineralized matrix prior to calcification, localizing at tissue interfaces such as cement lines and between collagen fibrils.
    While primarily produced by osteoblasts, it is also synthesized by brain and kidney cells. Osteopontin variants differ in molecular weight due to post-translational modifications.
    Functions include acting as a substrate for transglutaminase and involvement in cell adhesion, chemoattraction, and immunomodulation.

Target Information

Osteopontin is an RGD-containing glycoprotein that interacts with integrins and CD44 as major receptors. It is involved in bone mineralization, cell adhesion, migration, chronic inflammation, and transformation.
Proteolytic cleavage by thrombin and matrix metalloproteinases near the RGD sequence modulates OPN’s function and integrin binding.
Thrombin-cleaved fragments are overexpressed in malignant glial tumors, providing a survival niche and novel substrates for plasmin and cathepsin D.
