Target Information
Caspase 8 (CASP8)는 cysteine-aspartic acid protease (caspase) 계열의 단백질로, 세포사멸(apoptosis) 과정의 실행 단계에서 중심적인 역할을 합니다. Caspase들은 일반적으로 비활성 전구체 형태로 존재하며, 내부의 aspartic 잔기에서 절단되어 활성화된 효소로 전환됩니다. Caspase 8은 Fas 및 다양한 세포사멸 자극에 의해 유도되는 programmed cell death에 관여합니다. 또한 FADD 단백질과 상호작용하며, Huntington 병 환자의 뇌에서 불용성 형태로 검출된 바 있어 신경퇴행성 질환과의 관련성이 제시되었습니다. 활성화된 Caspase 8은 Caspase 9 등 다른 하위 caspase들을 활성화하여 세포사멸을 유도합니다. Caspase 8은 Jurkat 세포 및 편도(Tonsil)에서도 반응하며, 과발현 시 세포사멸을 유도합니다. 다양한 스플라이스 변이체가 보고되어 있으나, 모든 변이체의 전체 서열이 규명된 것은 아닙니다.
