Target Information

TMP21 is a ubiquitously expressed protein involved in vesicular targeting and protein transport.
It is part of the presenilin complex and modulates γ-secretase but not ε-secretase cleavage of amyloid precursor protein (APP).
The presenilin complex includes APH1, nicastrin, PEN2, and presenilin-1, which together cleave APP at γ- and ε-sites, leading to Abeta accumulation associated with Alzheimer’s disease.
Co-immunoprecipitation experiments indicate TMP21’s role in regulating this complex.
Suppression of TMP21 expression by siRNA increases γ-secretase activity and Abeta production, demonstrating TMP21’s modulatory function.
