Target Information

Huntington's disease (HD) is a neurodegenerative disorder characterized by loss of striatal neurons, caused by an expansion of a polyglutamine tract in the huntingtin protein. HAP1 was initially identified through two-hybrid library screening, showing binding correlation with huntingtin expansion. HAP1 interacts with cytoskeletal proteins such as dynactin and pericentriolar autoantigen protein 1, suggesting a role in vesicular trafficking or organelle transport. HAP1 is also involved in huntingtin-enhanced BDNF transport along microtubules, where attenuation leads to loss of neurotrophic support and neuronal toxicity, potentially contributing to disease pathogenesis. Several alternatively spliced isoforms have been described for HAP1.
