Target Information
ALAS-1은 glycine과 succinyl-CoA의 축합 반응을 촉매하여 delta-aminolevulinate, CO₂, CoA를 생성하는 핵 인코딩 미토콘드리아 효소입니다. heme 생합성 경로의 첫 번째 및 속도 제한 단계 조절에 관여하며, ALAS의 두 가지 아이소폼 중 하나입니다. Pyridoxal-5'-phosphate 의존성 효소로서, cytochrome 및 기타 hemoprotein의 heme 공급을 담당합니다. 간에서 heme에 의해 음성 피드백 조절을 받으며, ALAS1 유전자의 전사는 cAMP 및 호흡 해리로 자극되고, Phorbol ester와 insulin에 의해 억제됩니다. 또한 heme 생합성과 생체 시계의 상호 조절에 관여하여 circadian disorder 치료의 잠재적 표적이 될 수 있습니다. 급성 간부전에서는 ALAS1 발현이 감소하여 heme 대사 및 간 기능에 영향을 미칩니다.
