The eBio17B7 antibody reacts with mouse and rat IL-17A without recognition of IL-17F.
IL-17A is a CD4+ T cell-derived cytokine that promotes inflammatory responses and is elevated in autoimmune and inflammatory conditions such as rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.
Human IL-17A exists as glycosylated 20–30 kDa homodimers produced by activated CD4+ T cells. It enhances ICAM-1 expression in fibroblasts and stimulates epithelial, endothelial, and fibroblastic cells to secrete IL-6, IL-8, G-CSF, and PGE2. In the presence of IL-17A, fibroblasts can sustain proliferation of CD34+ progenitors and induce neutrophil maturation. Mouse, rat, and human IL-17A induce IL-6 secretion in mouse stromal cells, indicating cross-species receptor recognition.
IL-23-dependent IL-17A-producing CD4+ T cells (Th17 cells) represent a distinct subset regulated negatively by IFN-γ and IL-4.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested by intracellular staining and flow cytometry of restimulated, Th17-polarized splenocytes using the Fixation and Permeabilization Buffer Set (Product #88-8824-00). Recommended ≤0.25 µg/test (100 µL final volume; 10⁵–10⁸ cells/test).
Additional Protocols: Compatible with Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Excitation: 488 nm; Emission: 520 nm; Laser: Blue.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Interleukin-17A (IL-17A, CTLA-8) is a 32 kDa disulfide-linked homodimer (136 amino acids) that signals through IL-17R/CDw217. It regulates NF-κB and MAPK pathways, stimulates IL-6 and COX-2 expression, and enhances nitric oxide production. IL-17A is crucial in inflammatory and autoimmune responses.
