Product Specific Information
The eBio64DEC17 antibody reacts with human IL-17A and functions as a neutralizing antibody. IL-17A is a CD4+ T cell-derived cytokine involved in promoting inflammatory responses and is elevated in diseases such as rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection. It exists as a glycosylated 20–30 kDa homodimer and is produced by activated peripheral blood CD4+ T cells.
IL-17A enhances expression of ICAM-1 in fibroblasts and stimulates epithelial, endothelial, and fibroblastic cells to secrete IL-6, IL-8, G-CSF, and PGE2. It also supports proliferation of CD34+ hematopoietic progenitors and their maturation into neutrophils. Mouse, rat, and human IL-17A all induce IL-6 secretion in mouse stromal cells, indicating cross-species receptor recognition.
IL-23-dependent, IL-17A-producing CD4+ T cells (Th17 cells) represent a distinct subset of helper T cells, negatively regulated by Th1 and Th2 cytokines (e.g., IFN-γ, IL-4).
Intracellular staining with eBio64DEC17 identifies the same population as eBio64CAP17.
Applications Reported: Intracellular staining and flow cytometric analysis.
Applications Tested: Pre-titrated for intracellular staining of stimulated human peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test in 100 µL final volume.
Excitation/Emission: 488/520 nm (Blue Laser).
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Interleukin-17A (IL-17A, CTLA-8) is a 32 kDa disulfide-linked homodimer of 136 amino acids, part of the IL-17 family (IL-17A–F). It signals through IL-17R/CDw217 and regulates NF-κB and MAPK pathways, stimulating IL-6 and COX-2 expression and enhancing nitric oxide production. IL-17A plays a key role in inflammatory and autoimmune responses.
