Adenoviruses are DNA viruses widely distributed in nature and are frequent causes of acute upper respiratory tract infections such as the common cold. There are 49 known human adenovirus serotypes divided into six subgroups (A–E). They are associated with respiratory illness, conjunctivitis, gastroenteritis, and occasionally sexually transmitted diseases. Some types have oncogenic potential, though most cause self-limiting febrile illnesses involving inflammation of the conjunctivae and respiratory tract.

Adenoviruses can persist latently in tissues such as tonsils and adenoids and may be reactivated under immunosuppression (e.g., AIDS). Adenovirus vectors, especially those based on Ad5, are widely used in gene therapy research. The adenovirus capsid is composed of three main proteins: hexon, penton, and fiber. The hexon protein, a major coat component, forms 240 hexons per viral particle and is a homotrimer of a large (~900 residue) protein.
