Target Information

Adducins are heteromeric proteins composed of alpha, beta, and gamma subunits, encoded by distinct genes. They are membrane skeletal proteins involved in assembling the spectrin–actin network in erythrocytes and at cell–cell contact sites in epithelial tissues.

• Alpha and gamma adducins are ubiquitously expressed, while beta adducin is restricted to brain and hematopoietic tissues.
• Adducin was first purified from human erythrocytes as a heterodimer of alpha and beta subunits.
• Polymorphisms in these subunits have been linked to blood pressure regulation in animal models of hypertension.
• Heterodimers of alpha and gamma subunits have also been described.

Each subunit has two domains:

• Amino-terminal region: protease-resistant and globular.
• Carboxy-terminal region: protease-sensitive, contains phosphorylation sites for protein kinase C, a calmodulin-binding site, and is required for spectrin and actin association.

Alternatively spliced gamma adducin transcripts encoding different isoforms have been identified, though their specific functions remain unknown.
