Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into the cytoplasm of T cells and regulating the surface expression of the TCR complex. Activation through TCR involves the cytoplasmic tails of CD3 gamma, delta, epsilon, and zeta subunits—structurally related members of the immunoglobulin superfamily encoded by closely linked genes on chromosome 11.

CD3 subunits have long cytoplasmic tails that associate with signal transduction molecules via a double tyrosine-based motif. CD3 may play roles in TCR-induced growth arrest, cell survival, and proliferation. The CD3 antigen is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Reduced T lymphocyte percentages may occur in autoimmune diseases. Genes encoding CD3 epsilon, gamma, and delta are located on chromosome 11, and defects in CD3 genes are linked to CD3 immunodeficiency.
