Target Information
CHEK1 (CHK1) is a serine/threonine protein kinase involved in preventing DNA replication when integrity is compromised. It serves as a key mediator in the DNA damage-induced checkpoint network and ensures genomic stability under genotoxic stress.
Inhibition of CHK1 can abrogate the G2 or S checkpoint, leading to mitotic catastrophe and apoptosis in p53-deficient cancer cells, while normal cells remain arrested in the G1 phase. Therefore, CHK1 inhibitors can enhance the efficacy of DNA-damaging agents in cancer cells, making Chk1 an attractive therapeutic target, particularly since approximately 50% of human cancers are p53-deficient.
