Target Information
Epidermal Growth Factor (EGF)는 170 kDa 단백질 키나아제인 EGF 수용체(EGFR)에 결합하여 작용합니다. EGFR 활성화는 다양한 세포 반응 경로를 유도하며, EGF 결합 시 수용체는 동종 또는 이종 이합체를 형성합니다. 각 이합체는 SH2 도메인을 가진 다양한 효소 단백질을 모집하여 고유한 신호 경로를 개시합니다.
EGF는 신장, 뇌, 전립선, 침샘 등 다양한 조직에서 발현되며, 강력한 세포 증식 인자로 작용합니다. 인산화된 수용체는 GRB2 등의 어댑터 단백질을 모집하여 RAS-RAF-MEK-ERK, PI3K-AKT, PLCγ-PKC, STAT 경로를 포함한 주요 신호전달 경로를 활성화합니다. 또한 NF-κB 신호전달 경로 활성화에도 관여할 수 있음이 보고되었습니다.
EGF 유전자의 결함은 저마그네슘혈증 4형을 유발하며, 발현 이상은 특정 암의 성장 및 진행과 관련이 있습니다.
