Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3ε, a 20 kDa subunit of the TCR complex. The ε chain, along with γ and δ subunits, is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking via UCHT1 induces intracellular activation and proliferation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry of human peripheral blood cells (5 µL (0.4 µg)/test).
A test is defined as the antibody amount staining cells in 100 µL final volume (10⁵–10⁸ cells/test).

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability detection.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample prior to combining with cells
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells)

Technology:
NovaFluor conjugates utilize Phiton technology with engineered nucleic acid dye structures, providing optimized fluorescent signatures with reduced spillover and spread.

Target Information

The CD3 complex transduces antigen-recognition signals and regulates TCR surface expression. It consists of CD3γ, CD3δ, CD3ε, and CD3ζ subunits encoded by closely linked genes on chromosome 11. These subunits associate with cytoplasmic signal transduction molecules via tyrosine-based motifs. CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Defects in CD3 genes are associated with immunodeficiency.
