Product Specific Information

The monoclonal antibody eBio84-3C1 reacts with CD43, also known as leukosialin, galactoglycoprotein, and sialophorin.
CD43 is a sialomucin with both adhesive and anti-adhesive functions. It is expressed on most leukocytes except resting B lymphocytes, and its surface expression decreases upon activation. CD43 is involved in the activation of T cells, B cells, NK cells, and monocytes. The counter-receptor for CD43 is CD169/SIGLEC-1, expressed on macrophages.

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Recommended use: 5 µL (0.2 µg) per 100 µL cell sample (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample before mixing with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, using novel nucleic acid dye structures that minimize spectral spillover and spread.

Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with a high negative charge, expressed on most hematopoietic cells.
It functions as both a repulsive barrier and a promoter of leukocyte aggregation, depending on context. Through interactions with actin-binding proteins (ezrin, moesin), CD43 regulates T-cell morphology and cell-cell interactions during lymphocyte trafficking.
CD43 signaling enhances or counteracts LFA-1 adhesiveness, influences p53 protein induction, and can modulate tumor proliferation depending on cellular context.
Diseases associated with CD43 dysfunction include Wiskott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
