Product Specific Information

This BB7.2 monoclonal antibody recognizes the human major histocompatibility complex (MHC) class I molecule HLA-A2, a member of the HLA-A family. MHC class I consists of a single alpha chain with three domains, associated with beta 2-microglobulin, and is expressed on all nucleated cells.

The BB7.2 antibody exhibits blocking activity and also recognizes HLA-A28.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometry using normal human peripheral blood cells. Recommended use is 5 µL (0.6 µg) per test in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

NovaFluor dyes are incompatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

This product ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance. Add 5 µL per sample to antibody cocktails prior to combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized spectral performance with minimal spillover and spread.

Target Information

HLA-A is a class I molecule consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain (~45 kDa) contains 8 exons:

  • Exon 1: leader peptide
  • Exons 2–3: α1 and α2 domains (peptide binding)
  • Exon 4: α3 domain
  • Exon 5: transmembrane region
  • Exons 6–7: cytoplasmic tail

Polymorphisms in exons 2 and 3 determine peptide-binding specificity. HLA-A typing is routinely used in bone marrow and kidney transplantation. HLA/MHC antibodies are also key tools in immunopeptidomics for neoantigen identification via LC-MS/MS.
