Product Specific Information

The 10C4 monoclonal antibody reacts with mouse and rat Bcl-2, also known as B cell lymphoma 2. Bcl-2 is the founding member of the Bcl-2 family of proteins that regulate cell death and survival. Members include pro-apoptotic proteins (BAD, Bax, Bim) and anti-apoptotic proteins (Bcl-2, Bcl-xL, Mcl-1).

Bcl-2 inhibits apoptosis by preventing cytochrome c release from mitochondria. Overexpression or mutation of Bcl-2 has been associated with various cancers. In thymocytes, Bcl-2 expression varies by developmental stage.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested with mouse splenocytes and thymocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).

Usage: 5 µL (0.25 µg)/test, defined as staining a sample in 100 µL final volume.
Cell number: 10⁵–10⁸ cells/test (empirically determined).

eFluor® 660 replaces Alexa Fluor® 647, emits at 659 nm, excited by red laser (633 nm).
Excitation: 633–647 nm; Emission: 668 nm; Laser: Red.
Post-manufacturing filtration: 0.2 µm.

Target Information

BCL-2 regulates apoptosis by inhibiting or promoting cell death. It forms dimers with other family members (e.g., Bag1, Bax, Bak) to modulate apoptotic signaling.
Overexpression of BCL-2, often due to translocation, is linked to follicular lymphoma and other cancers.
Two transcript variants exist, differing in C-terminal ends. High BCL-2 expression is typical in neoplastic germinal centers but absent in normal ones.
