Target Information

Phosphofructokinase (PFK) is a major regulatory enzyme that catalyzes sugar metabolism, pivotal in energy production for normal cell function. In humans, three loci control PFK expression: M (muscle), L (liver), and P (platelet) type subunits, variably expressed across tissues.
PFK forms tetramers from combinations of M and L subunits, resulting in five possible isozymes. It is activated by ADP, AMP, or fructose bisphosphate and inhibited by ATP or citrate.
Deficiency in PFK can cause episodic illness, characterized by dark urine after strenuous exercise. Elevated red cell PFK levels are observed in individuals with Down syndrome.
