Target Information
Focal Adhesion Kinase (FAK) is a 125 kDa non-receptor protein tyrosine kinase that functions as a substrate for Src and plays a crucial role in integrin signaling. It regulates cell spreading, differentiation, migration, apoptosis, and the G1–S phase transition.
FAK contains a central catalytic domain and a C-terminal tail that localizes it to focal adhesions, sites of cell attachment to the extracellular matrix via integrin receptors. Increased FAK tyrosine phosphorylation occurs upon integrin engagement with fibronectin.
In NIH3T3 fibroblasts, adhesion to fibronectin promotes association of Grb2 and c-Src with FAK, leading to activation of ERK2 MAP kinase. In v-Src-transformed NIH3T3 cells, this association occurs independently of adhesion. The Grb2 SH2 domain binds directly to tyrosine-phosphorylated FAK at Tyr925.
Several transcript variants encoding different isoforms have been identified, with three full-length forms characterized.
