Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. Upon activation, p53 regulates the transcription of target genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 contains an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation at Ser15 is critical for p53 activation and stabilization, while phosphorylation at Ser392 has been associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells and functions as a natural anticancer agent. It is expressed at low levels in normal cells but is elevated in many transformed or malignant cell lines. Mutations in the TP53 gene are common in human cancers and can result in loss of tumor suppressor function. Germline TP53 mutations are also linked to hereditary cancer syndromes such as Li-Fraumeni syndrome.
