• For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with a high negative charge, expressed on the surface of most hematopoietic cells.
It contributes to a repulsive barrier that interferes with cellular adhesion, but can also promote leukocyte aggregation.
Through interactions with actin-binding proteins (ezrin and moesin), CD43 regulates T-cell morphology and cell-cell interactions during lymphocyte trafficking.
CD43 signaling enhances LFA-1 adhesiveness while counteracting its induction via other receptors.
Expression of CD43 can induce functionally active tumor suppressor p53 protein; however, in the absence of p53 and ARF, CD43 may promote tumor proliferation and viability.
Diseases associated with CD43 dysfunction include Wiskott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
