Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDKs) and is transcriptionally activated by p53.
It plays a critical role in the cellular response to genotoxic stress and acts as a major transcriptional target of p53.
In the nucleus, p21 binds and inhibits CDK1 and CDK2, blocking the cell cycle transition from G1 to S phase or G2 to mitosis after DNA damage, allowing DNA repair.
In the cytoplasm, p21 exhibits anti-apoptotic effects by inhibiting caspase 3 and apoptotic kinases ASK1 and JNK.
Thus, p21 has a dual function in carcinogenesis, acting as both a tumor suppressor and an oncogene depending on context.
