Target Information
Protein disulfide isomerase (PDI) is a multifunctional protein that catalyzes the formation, breakage, and rearrangement of disulfide bonds.
At the cell surface, PDI acts as a reductase that cleaves disulfide bonds of proteins attached to the cell, potentially causing structural modifications of exofacial proteins.
Inside the cell, it facilitates disulfide bond formation and rearrangement in nascent proteins.
At high concentrations, PDI functions as a chaperone preventing aggregation of misfolded proteins, while at low concentrations, it can promote aggregation (anti-chaperone activity).
PDI may also participate with other chaperones in structural modification of the TG precursor in hormone biogenesis and serves as a structural subunit of enzymes such as prolyl 4-hydroxylase and microsomal triacylglycerol transfer protein (MTTP).
It also acts as a receptor for LGALS9, retaining P4HB at the cell surface of Th2 T helper cells, which enhances disulfide reductase activity and cell migration.
