Product Specific Information

The 236A/E7 antibody reacts with human Foxp3 protein, also known as FORKHEAD BOX P3, SCURFIN, and JM2. Foxp3 (49–55 kDa) is a member of the forkhead/winged-helix family of transcriptional regulators and is the gene defective in scurfy (sf) mice. High expression of Foxp3 mRNA is observed in CD4+CD25+ regulatory T cells (Treg), and ectopic expression in CD4+CD25− cells imparts a Treg phenotype.

Intracellular staining and flow cytometric analysis using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) show staining of the CD4+CD25^bright population. The epitope recognized by 236A/E7 differs from that of PCH101 (Product #72-5776-40). This antibody also recognizes rhesus macaque, sooty mangabey, and cynomolgus macaque.

Intracellular staining followed by flow cytometric analysis and immunohistochemistry (paraffin and frozen sections).

Applications Tested:
Tested by intracellular staining and flow cytometric analysis of normal human peripheral blood cells using the Foxp3/Transcription Factor Buffer Set (Product #00-5523-00). Refer to BestProtocols® for “Staining Intracellular Antigens for Flow Cytometry” (Protocol B). Recommended use ≤0.5 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is expressed highly in CD25+CD4+ regulatory T cells and minimally in CD4+/CD25− cells, absent in CD4−/CD8+ T cells. FOXP3 acts as a master regulatory gene and specific marker of Treg cells. Mutations cause the scurfy phenotype in mice and IPEX syndrome in humans (immune dysfunction, polyendocrinopathy, enteropathy, X-linked). FOXP3 expression is p53-regulated in human breast and colon cancer cells.
