이 항체는 rabbit serum에서 순차적인 epitope-specific chromatography로 정제되었습니다. 비인산화 EGFR에 반응하는 항체를 제거하기 위해 비인산화 펩타이드로 음성 예흡착 과정을 거쳤으며, 최종 제품은 Tyr1068에서 인산화된 EGFR 유래 펩타이드로 친화 크로마토그래피를 통해 생산되었습니다. Western blot의 양성 대조군으로는 EGF 처리 전후의 A431 세포가 사용되었습니다.
Target Information
EGFR (Epidermal Growth Factor Receptor)은 다양한 성장 인자에 반응하여 신호를 전달하는 수용체 티로신 키나아제입니다. 과발현은 여러 암종과 관련이 있으며, 다양한 생물학적 및 화학적 치료제의 표적이 됩니다. EGFR 유전자는 인간 7번 염색체에 위치하며, HER/ERbB 단백질 패밀리(ERbB2, ERbB3, ERbB4 포함)에 속합니다. EGF나 TGFα와 같은 리간드가 결합하면 EGFR은 이합체화되고, 세포질 도메인의 티로신 잔기가 자가 인산화되어 신호 전달을 유도합니다. EGFR 과발현은 두경부, 뇌, 방광, 위, 유방, 폐, 자궁내막, 자궁경부, 난소 등 다양한 종양에서 관찰됩니다.
