Target Information
PSD-93, also known as chapsyn-110, is a plasma membrane-associated protein found in synaptic junctions. It is uniquely expressed in Purkinje neuron cell bodies and dendrites. PSD-93 contains three PDZ domains (~90 amino acids each), one SH3 domain, and a carboxyl-terminal guanylate kinase homology (GuK) domain that is enzymatically inactive.
PDZ-domain interactions are believed to play a role in receptor and channel clustering contributing to neuronal plasticity. PSD-93 participates in clustering of NMDA receptors and shaker-type potassium channels at the synaptic membrane.
These proteins share a C-terminal sequence motif (T/SXV), which mediates PDZ-domain binding. Neuronal nitric oxide synthase (nNOS), lacking this motif but containing its own PDZ domain, can interact with PSD-93 via a pseudo-homotypic PDZ-PDZ interaction.
