Target Information

The mediator complex is a multi-protein transcriptional co-activator expressed ubiquitously in eukaryotes and required for RNA polymerase II transcription induction. MED28 (Magicin) is one of the proteins in this complex and is expressed in many cell lines and tissues. Down-regulation of MED28 in NIH3T3 cells induces genes associated with smooth muscle cell differentiation, while over-expression represses SMC gene expression. MED28 can form a ternary complex with Grb2 and Merlin, suggesting a role in Merlin’s tumor suppressive activity. MED28 has also been identified as an HIV dependency factor (HDF), implying its potential as a drug target in HIV treatment. At least two isoforms of MED28 are known.
