Product Specific Information
- For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
NPM1 (Nucleophosmin 1, also known as B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in:
- Ribosome assembly and transport
- Cytoplasmic/nuclear trafficking
- Regulation of DNA polymerase α activity
- Centrosome duplication and p53 regulation
NPM1 shuttles between the nucleus, cytoplasm, and nucleolus, chaperoning core histones and preventing protein aggregation. It binds nucleic acids, protects enzymes during thermal denaturation, and assists in protein renaturation. NPM1 is associated with centrosome duplication as a substrate of CDK2/cyclin E. It also participates in tumor-related fusion proteins such as NPM-ALK, NPM-RAR, and NPM-MLF1, acting as an interface for oligomerization and oncogenic conversion. Dysfunction of NPM1 is linked to diseases such as acute myeloid leukemia and lymphomatoid papulosis.
