Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells. CD20 expression coincides with surface IgM expression and regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 is associated with lipid rafts, and its interaction depends on extracellular signals, conformational changes, and BCR aggregation. Upon receptor ligation, CD20 and BCR colocalize transiently before BCR endocytosis, while CD20 remains at the surface. Due to its stable surface expression, CD20 serves as a key therapeutic target for B-cell depletion in malignancies. Dysfunctions in CD20 (MS4A1) are linked to common variable immune deficiency.
