Target Information
tPA converts the abundant but inactive zymogen plasminogen to plasmin by hydrolyzing a single Arg-Val bond in plasminogen. By controlling plasmin-mediated proteolysis, it plays an important role in tissue remodeling, degradation, cell migration, and other physiological and pathological events.
tPA is a heterodimer of chain A and chain B held by a disulfide bond and binds to fibrin with high affinity, increasing catalytic efficiency by 100–1000-fold. Binding to heparin, laminin, and fibronectin enhances plasminogen activation.
It also interacts with mannose receptor and LRP1, which are involved in tPA clearance, as well as annexin II and cytokeratin 8. Binding to vascular smooth muscle cells reduces inhibition by PAI-1 and stimulates plasminogen activation.
