Target Information

Tumor necrosis factor (TNF) induced signaling is mediated through association of TNF receptor (TNFR) with adaptor proteins such as TNF receptor associated factors (TRAFs). TRAFs are cytoplasmic adapter proteins that mediate signal transduction from TNF-receptor superfamily members (e.g., RANK, CD30, CD40) and the interleukin-1 receptor.
The carboxy-terminal region of TRAFs is essential for self-association and receptor interaction following ligand-induced oligomerization. Six TRAFs (TRAF1–TRAF6) have been identified.
TRAF5 (558 amino acids) is involved in NF-κB and c-Jun N-terminal kinase activation by TNF receptor superfamily members including CD27, CD30, CD40, and lymphotoxin-beta receptor. Disruption of the TRAF5 gene causes defects in CD40-CD27 mediated lymphocyte activation.
