Target Information
Retinoic acid-inducible gene I (RIG-I) is a pattern recognition receptor (PRR) that detects viral double-stranded RNA (dsRNA) and activates the innate immune response. Along with MDA5, RIG-I plays a critical role in antiviral defense, recognizing short dsRNAs lacking a 5′-triphosphate end. Activation of RIG-I leads to signaling through IPS-1, which triggers transcription factors NF-kappaB and IRF-3, resulting in apoptosis, cytokine signaling, and inflammation.
RIG-I is essential for immune signaling against influenza A/B, human respiratory syncytial virus, paramyxoviruses, Japanese encephalitis virus, and West Nile virus. MicroRNA-146a negatively regulates RIG-I-dependent antiviral responses by targeting TRAF6, IRAK1, and IRAK2. Recent studies suggest RIG-I also detects cytosolic DNA via RNA polymerase III activity.
