Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
    Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

  • Western Blot: Use 1–2 µg/mL for 60 minutes at room temperature.

Target Information

CD47 (Integrin-associated protein, IAP) is a ubiquitously expressed cell surface transmembrane glycoprotein that interacts with several integrins and regulates their functions. Engagement of CD47 by soluble ligands or counter receptors modulates various signaling pathways, including activation of heterotrimeric G proteins. Functionally, CD47 is involved in increasing intracellular calcium concentration upon cell adhesion to the extracellular matrix.

CD47 also serves as a receptor for the C-terminal cell-binding domain of thrombospondin, playing a role in membrane transport and signal transduction. It has broad tissue distribution but reduced expression on Rh erythrocytes. CD47 acts as a ligand for CD172a (signal regulatory protein alpha, SIRPα), an immune inhibitory receptor on macrophages, preventing phagocytosis of CD47-positive cells. The CD47–CD172a system affects cell migration, B cell adhesion, T cell activation, modulates chondrocyte responses to mechanical signals, and promotes neuronal development, being especially abundant in synapse-rich regions of the brain and retina.
