Product Specific Information

The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is essential for assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells.

Binding of 145-2C11 to TCR initiates intracellular signaling leading to activation, proliferation, or apoptosis. Commonly used as a phenotypic marker for mouse T cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes (≤1.0 µg/test, 100 µL volume)

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer: Required for optimal staining. Add 5 µL per sample (10³–10⁸ cells). For single-color controls, use 5 µL per 100 µL cell sample.

Target Information

The CD3 subunit complex transduces antigen-recognition signals and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded on chromosome 11. CD3 is expressed on most T cells, thymocytes, and Purkinje cells, but not on B or NK cells.

Defects in CD3 genes are associated with CD3 immunodeficiency. CD3 plays roles in TCR-induced growth arrest, survival, and proliferation.
