Product Specific Information

This MR6F3 antibody recognizes mouse CD206 (Macrophage Mannose Receptor, MMR or MRC1), a 175-kDa type I membrane glycoprotein expressed in macrophages, dendritic cells, and endothelial cells. CD206 belongs to the C-type lectin family and mediates phagocytic and endocytic uptake of microbial antigens, playing a key role in immune defense and regulation.

CD206 expression is induced by IL-4, IL-13, M-CSF, IL-6, IL-10, and glucocorticoids, while TNF-α, IFN-γ, TGF-β, and LPS down-regulate it. It is a marker for M2 macrophages. Intracellular staining is recommended due to limited surface expression.

Applications Reported: Flow cytometric analysis
Applications Tested: Intracellular staining and flow cytometry using Intracellular Fixation and Permeabilization Buffer Set (Product #88-8824-00).
Use ≤0.25 µg/test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Fluorochrome Information:
PE-eFluor 610 — Excitation: 488–561 nm, Emission: 607 nm. Recommended filter: 610/20 band pass (equivalent to PE-Texas Red).
Light sensitive; protect from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

Target Information

CD206 (Mannose receptor, MRC1) is a 175 kDa transmembrane protein in the pattern recognition receptor family. Expressed mainly in macrophages, dendritic cells, endothelial cells, and sperm cells, it plays a role in innate and adaptive immunity. It mediates uptake of macromolecules such as modified LDLs, AGEs, and amyloid β-protein.

CD206 dysfunction is linked to diseases including atherosclerosis, Alzheimer’s disease, leprosy, and Gaucher’s disease. Isoforms 1 and 2 mediate endocytosis, while isoform 3 lacks this function and can inhibit others through a dominant-negative effect.
