Target Information

PTEN is a tumor suppressor frequently mutated in various cancers. It encodes a phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase, containing a tensin-like domain and a catalytic domain similar to dual-specificity protein tyrosine phosphatases.
Unlike most tyrosine phosphatases, PTEN preferentially dephosphorylates phosphoinositide substrates, negatively regulating intracellular levels of phosphatidylinositol-3,4,5-trisphosphate and AKT/PKB signaling pathway.
PTEN modulates the AKT-mTOR pathway, influencing neuronal integration, dendritic development, and synapse formation during adult neurogenesis. It also acts as a negative regulator of insulin signaling and glucose metabolism in adipose tissue.
The nuclear monoubiquitinated form of PTEN shows higher apoptotic potential, while the cytoplasmic nonubiquitinated form exhibits reduced tumor suppressive ability.
