Target Information
MLH1 is a DNA mismatch repair protein. The repair of mismatched DNA is essential for maintaining the integrity of genetic information over time. Alterations of microsatellite repeats, resulting from strand misalignment during DNA replication, lead to microsatellite instability (MSI). Defects in DNA repair pathways have been linked to human carcinogenesis. The importance of mismatch repair genes became evident with the identification of the genetic basis for hereditary nonpolyposis colon cancer (HNPC). MSH2 is involved in the initial recognition of mismatch nucleotides during replication repair. After MSH2 binds to mismatched DNA, it forms a heterodimer with MLH1 and PMSH, facilitating later steps in mismatch repair.
