Target Information
Phosphoinositide-specific phospholipase C (PLC) plays a crucial role in receptor-mediated signal transduction through the generation of the second messengers inositol 1,4,5-triphosphate and diacylglycerol from phosphatidylinositol 4,5-bisphosphate.
There are many mammalian PLC isozymes, including PLCβ1, PLCβ2, PLCβ3, PLCβ4, PLCγ1, PLCγ2, PLCδ1, PLCδ2, and PLCε.
PLCδ exists as four isoforms. PLCδ1, a calcium signal amplifier, is activated by an atypical GTP-binding protein and acts as an effector in oxytocin receptor and α1B-adrenoreceptor signaling. It is highly expressed in brain, heart, lung, and testis. PLCδ is abnormally accumulated in brains with Alzheimer’s disease, suggesting a role in AD pathology.
PLCδ2 is markedly expressed in type II intestinal metaplasia and adenocarcinoma. When expressed in type I intestinal metaplasia, it is generally benign but may evolve toward neoplastic transformation, suggesting PLCδ2 as a potential marker of gastric malignant transformation.
