• Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

Amyloid beta peptide (Abeta/Beta-amyloid) is the major constituent of amyloid plaques in the brains of individuals afflicted with Alzheimer’s disease. Abeta peptide is 40–43 amino acids long and generated from the beta-amyloid precursor protein (beta APP) in a two-step process.
Beta APP is cleaved by beta-secretase (BACE) and gamma-secretase, producing the amyloid beta peptide. Gamma-secretase is a multi-subunit complex containing presenilin-1 and -2 as central components, and nicastrin as an associated transmembrane glycoprotein that modulates peptide production.
Abeta is an extracellular filamentous protein component of amyloid cores and neuritic plaques, also found in neurofibrillary tangles. Beta amyloid deposits are characteristic of Alzheimer’s disease and are also observed in Lewy body dementia, Down’s syndrome, Dutch-type amyloidosis, cerebral amyloid angiopathy, and the Guam Parkinson-Dementia complex.
