Product Specific Information

Description:
The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells excluding circulating erythrocytes and platelets. The cytoplasmic portion of CD45 has tyrosine phosphatase activity and plays an important role in lymphocyte activation.

Applications Reported:
Validated for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test. A test is defined as the amount of antibody that stains cells in a 100 µL final volume, with 10⁵–10⁸ cells/test.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. Absent in non-hematopoietic tissues, CD45 is critical for lymphocyte development and antigen signaling. It regulates Src-family kinases, JAK kinase activity, and integrin-mediated signaling, influencing immune responses, apoptosis, and cytokine signaling. Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
