Target Information
Estrogen Receptor beta (ER beta, NR3A2) protein is approximately 55 kDa, though multiple isoforms ranging from 36–59 kDa have been described. Human ER beta was initially cloned and characterized from testis.
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor acts as a ligand-activated transcription factor that, upon binding to estrogen, undergoes conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA.
ERs regulate gene transcription positively or negatively via coactivator or corepressor recruitment. Two forms exist: ER alpha (ESR1) and ER beta (ESR2). ER beta binds estrogens with similar affinity to ER alpha and can inhibit ER alpha activity in reproductive tissues.
ER beta is widely expressed and functions as a tumor suppressor, relevant to diseases such as prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain disease, and cardiovascular disease risk.
