- Immunohistochemistry (PFA fixed):
Incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at room temperature for 20 min.
Target Information
CD14 is a 55 kDa GPI-anchored glycoprotein expressed on the surface of mature monocytes, macrophages, and neutrophils.
It serves as a multifunctional lipopolysaccharide receptor and is released to the serum both as a secreted and enzymatically cleaved GPI-anchored form.
CD14 binds lipopolysaccharide in a reaction catalyzed by LBP (lipopolysaccharide-binding protein), an acute phase serum protein.
The soluble sCD14 can discriminate slight structural differences between lipopolysaccharides and plays a role in neutralization of serum lipopolysaccharides.
Additionally, CD14 binds apoptotic cells and influences allergic, inflammatory, and infectious processes.
Alternative splicing results in multiple transcript variants encoding the same CD14 isoform.
Diseases associated with CD14 dysfunction include Mycobacterium chelonae infection and croup.
