Target Information

CXXC4 is a zinc finger protein that binds directly to the PDZ domain of DVL1, a protein involved in the Wnt signaling pathway, through its C-terminal region. The interaction between CXXC4 and DVL1 competes with DVL1-AXIN binding, although DVL1 has higher affinity for AXIN. In mouse fibroblasts, CXXC4 suppresses Wnt3a-induced beta-catenin activation and inhibits Wnt3a-dependent TCF4 activation, indicating its role as a negative regulator in the Wnt signaling pathway.
Decreased CXXC4 expression has been associated with kidney cancer metastasis, enhanced cell proliferation, reduced apoptosis in response to cancer drugs, and upregulation of genes related to proliferation, invasion, and survival, suggesting its critical role in tumor progression.
