  • Positive Control: HeLa, U87, or Ramos cells
  • Cellular Location: Cytoplasm and nucleus. IRF3 shuttles between cytoplasmic and nuclear compartments; interaction with CREBBP prevents export to the cytoplasm, though export is typically predominant. When activated, IRF3 translocates to the nucleus.

Target Information

IRF3 (Interferon Regulatory Factor 3) is a key transcriptional regulator of type I interferon (IFN)-dependent immune responses. It plays a crucial role in innate immunity against DNA and RNA viruses by regulating transcription of IFN-alpha, IFN-beta, and interferon-stimulated genes (ISG) through binding to ISRE elements in their promoters.

IRF3 is inactive in the cytoplasm of uninfected cells. Upon viral infection or stimulation via double-stranded RNA or toll-like receptor pathways, it becomes phosphorylated by IKBKE and TBK1 kinases, leading to dimerization, nuclear localization, and complex formation with CREB binding protein (CBP) to activate IFN and ISG transcription. Mutations in IRF3 can cause acute infection-induced encephalopathy (herpes-specific type 7).
