Target Information
Lipolytic enzymes are essential for mobilization of fatty acids from triglyceride stores in adipose tissue. Dysfunctional lipolysis affects energy homeostasis and may contribute to obesity and insulin resistance.
ATGL (Adipose Triglyceride Lipase) catalyzes the initial step of triglyceride hydrolysis and is highly expressed in adipose tissue of mice and humans. It exhibits high substrate specificity for triacylglycerol and associates with lipid droplets. Inhibition of ATGL reduces total adipose acyl-hydrolase activity, indicating its cooperative role with hormone-sensitive lipase (HSL) in triglyceride catabolism.
