Target Information
CD22 (BL-CAM) is a type 1 integral membrane glycoprotein (130–140 kDa) expressed on B-lymphocytes, both in cytoplasm and cell membrane. It appears early in B-cell differentiation, similar to CD19 antigen.
Membrane expression is limited to mature B cells (CD22+) and plasma cells (CD22−), useful for phenotyping mature leukemias. Strongly expressed in hairy cell leukemia, diffuse large B-cell lymphoma, and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
CD22 binds preferentially to α2,6-linked sialic acid, regulating B-cell receptor signaling through interactions with Src family kinases and phosphatases.
