Target Information

This gene encodes a member of the F-box protein family characterized by an approximately 40 amino acid motif, the F-box. F-box proteins are subunits of the SCF ubiquitin ligase complex (SKP1-cullin-F-box), functioning in phosphorylation-dependent ubiquitination.
F-box proteins are categorized into three classes:

1. Fbws containing WD-40 domains
2. Fbls with leucine-rich repeats
3. Fbxs with various protein-protein interaction modules or no recognizable motifs

The BTRC protein belongs to the Fbws class and contains multiple WD-40 repeats. It is homologous to Xenopus bTrCP1, yeast Met30, Neurospora Scon2, and Drosophila Slimb proteins. BTRC interacts with HIV-1 Vpu, linking CD4 to the proteolytic machinery, and associates with phosphorylated IkappaBalpha and beta-catenin destruction motifs. It functions in transcriptional regulation by activating NF-kappaB and inhibiting beta-catenin pathways.
